Abstract Background Currently, only a few studies have described the general characteristics of patients with primary Sjögren's syndrome (pSS) who tested negatives for anti-SSA and anti-SSB antibodies. We aimed to further investigate the clinical characteristics of these patients in a large sample. Methods Data from patients with pSS who were treated at a tertiary hospital in China between 2013 and 2022 were retrospectively analyzed. Clinical characteristics of the patients were compared between those with and without anti-SSA and anti-SSB antibody negativity. Factors associated with anti-SSA and anti-SSB negativity were identified by logistic regression analysis. Results Overall, 934 patients with pSS were included in this study, among whom 299 (32.0%) tested negative for anti-SSA and anti-SSB antibodies. Compared with patients testing positive for anti-SSA or anti-SSB antibodies, that testing negative for the two antibodies had a lower proportion of females (75.3% vs. 90.6%, p < 0.001) and thrombocytopenia (6.7% vs. 13.6%, p = 0.002), but a higher proportion of abnormal Schirmer I tests (96.0% vs. 89.1%, p = 0.001) and interstitial lung disease (ILD) (59.2% vs. 28.8%, p = 0.001). Anti-SSA and anti-SSB negativity was positively associated with male sex (odds ratio [OR] = 1.86, 95% confidence interval [CI]: 1.05, 3.31), abnormal Schirmer I tests (OR = 2.85, 95% CI: 1.24, 6.53), and ILD (OR = 2.54, 95% CI: 1.67, 3.85). However, it was negatively related to thrombocytopenia (OR = 0.47, 95% CI: 0.24, 0.95). Conclusion Approximately one third of pSS patients had anti-SSA and anti-SSB negativity. pSS patients testing negative for anti-SSA and anti-SSB showed a higher risk of abnormal Schirmer I tests and ILD, but a lower risk of thrombocytopenia.

Objective: To investigate the effect of acetyl-CoA carboxylase 1 (ACC1) knockdown on the migration of esophageal squamous cell carcinoma (ESCC) KYSE-450 cell and underlying mechanism. Methods: Lentiviral transfection was conducted to establish sh-NC control cell and ACC1 knocking down cell (sh-ACC1). Human siRNA HSP27 and control were transfected by Lipo2000 to get si-HSP27 and si-NC. The selective acetyltransferase P300/CBP inhibitor C646 was used to inhibit histone acetylation and DMSO was used as vehicle control. Transwell assay was performed to detect cell migration. The expression of HSP27 mRNA was examined by reverse transcription-quantitative real-time polymerase chain reaction (RT-qPCR) and the expressions of ACC1, H3K9ac, HSP27 and epithelial-mesenchymal transition-related proteins E-cadherin and Vimentin were detected by western blot. Results: The expression level of ACC1 in sh-NC group was higher than that in sh-ACC1 group (P<0.01). The number of cell migration in sh-NC group was (159.00±24.38), lower than (361.80±26.81) in sh-ACC1 group (P<0.01). The protein expression levels of E-cadherin and Vimentin in sh-NC group were statistically significant compared with sh-AAC1 group (P<0.05). The migrated cell number in sh-NC+ si-NC group was (189.20±16.02), lower than (371.60±38.40) in sh-ACC1+ si-NC group (P<0.01). The migrated cell number in sh-NC+ si-NC group was higher than that in sh-NC+ si-HSP27 group (152.40±24.30, P<0.01), and the migrated cell number in sh-ACC1+ si-NC group was higher than that in sh-ACC1+ si-HSP27 group (P<0.01). The protein expression levels of E-cadherin and Vimentin in sh-NC+ si-NC group were significantly different from those in sh-ACC1+ si-NC and sh-NC+ si-HSP27 groups (P<0.01). The protein expression levels of E-cadherin and Vimentin in sh-ACC1+ si-NC group were significantly different from those in sh-ACC1+ si-HSP27 group (P<0.01). After 24 h treatment with C646 at 20 μmmo/L, the migrated cell number in sh-NC+ DMSO group was (190.80±11.95), lower than (395.80±17.10) in sh-ACC1+ DMSO group (P<0.01). The migrated cell number in sh-NC+ DMSO group was lower than that in sh-NC+ C646 group (256.20±23.32, P<0.01). The migrated cell number in sh-ACC1+ DMSO group was higher than that in sh-ACC1+ C646 group (87.80±11.23, P<0.01). The protein expressions of H3K9ac, HSP27, E-cadherin and Vimentin in sh-NC+ DMSO group were significantly different from those in sh-ACC1+ DMSO group and sh-NC+ C646 group (P<0.01). The protein expression levels of H3K9ac, HSP27, E-cadherin and Vimentin in sh-ACC1+ DMSO group were significantly different from those in sh-ACC1+ C646 group (P<0.01). Conclusion: Knockdown of ACC1 promotes the migration of KYSE-450 cell by up-regulating HSP27 and increasing histone acetylation.

[Abstract] Objective To study the correlation between follicular helper T (TFH) cells with the cellular immunity indices and the percentage of follicular cytotoxic T (TFC) cells in peripheral blood of human immunodeficiency virus-1 (HIV-1) infected patients and explore the relationship between TFH cells and the acquired immunodeficiency symdrome (AIDS) disease progression. Methods Seventeen health controls and 65 HIV-1 infected individuals were enrolled in this study, and the HIV-1 infected patients included 48 untreated HIV-1 positive patients and 17 treated with highly active antiretroviral therapy, the 48 HIV-1 infected patients consisted of 9 patients with CD4+ T cells ≤200/μl, 19 with 200/μl 350/μl. The expression of CXCR5 on CD4+ T cells was detected by flow cytometry and its correlation with CD4/CD8 ratio and HIV-1 RNA viral load. We also analyzed the correlation between the percentage and absolute number of TFH cells and TFC cells. Results Compared with healthy controls, both the absolute cell numbers [(60.23±27.62)/μl vs. (207.53±51.50)/μl, U=0, P<0.0001] and frequency [(18.83±4.55)% vs. (22.06±5.35)%, U=242.00, P=0.0124] of TFH cells were significantly reduced in untreated HIV-1 infected patients; The absolute number of TFH cells in the antiretroviral therapy group was significantly higher than that in untreated HIV-1 infected patients [(135.05±60.77)/μl vs. (60.23±27.62)/μl, U=108.00, P=0.0082]. The frequency of TFH cells in untreated HIV-1 infected patients were negatively correlated with CD4/CD8 ratio (r=–0.338, P=0.019), positively correlated with viral load (r=0.392, P=0.006). Both the absolute cell numbers (r=0.408, P=0.004), and frequency (r=0.363, P=0.011) of TFH cells were positively correlated with the frequency of TFC cells. The percentage of TFH cells decreased with the decrease of CD4+ T cells and the number of TFH cells increased with the decrease of CD4+ T cells in untreated HIV-1 infected patients. Conclusions TFH cells may be associated with active replication of the HIV-1 and disease progression. The decrease of TFH absolute cell numbers in patients with HIV-1 infection may be related to the cytotoxicity of TFC cells.

OBJECTIVES: To introduce a new laparoscopic splenectomy (LS) approach. METHODS: Sixteen patients underwent LS with general anaesthesia and carbon dioxide pneumoperitoneum. The details of the surgery are as follows: 1. The omentum was incised along the greater curvature and retracted as much as possible to expose the pancreatic body and tail. 2. The right arteriovenous root in the gastric omentum was ligated to sufficiently expose the pancreatic body and tail. 3. The pancreatic capsula was opened along the inferior margin of the pancreatic tail, elevated and separated until the superior margin of the pancreas was grasped. The entire splenic pedicle was retracted using a string. The branching blood vessels in the splenic hilus were ligated using clamps and separated. The splenogastric and splenophrenic ligaments were transected proximally using an ultrasonic knife, and the thick short gastric blood vessels were clamped. This procedure allows complete exposure of the area above the pancreatic tail where the splenic hilus is located. The splenoportal vasculature was suspended using a 7-0 silk suture to easily manipulate this tissue. The splenic portal vessels were dissected using an ultrasonic knife, and the portal vessels were isolated individually using vascular clamps and transected. The splenogastric and lienorenal ligaments were also transected. The spleen was then placed into a bag, and the surgical port was slightly enlarged. Finally, the spleen was sectioned for removal. RESULTS: Fifteen surgeries were successfully performed from March 2015 to January 2016. One patient underwent laparotomy. No patients developed postoperative intra-abdominal haemorrhage or infection. One patient developed subcutaneous emphysema, and one developed a wound infection. No deaths occurred. CONCLUSIONS: Active exposure of the area dorsal to the pancreatic tail is a safe and simple splenectomy method.

Objective To explore the clinical application effects of the logistics robot as innovative mode in consumables management in operating room.Methods Clinical application experiment using self-designed medical logistics robot was conducted.During single point distribution,indicators including the average time/trip of logistics robots,the error rate of transporting goods,replacing the workload of the distributor,the incidence of cost loss,the right rate of avoiding pedestrians,the right rate of avoiding obstacles,the number of being lost,and the speed of operational reaction were recorded.The time of picking(T1),the delivery time(T2),and the check time(T3) of test between logistics robot and manual operation were compared.Results Experiment results were obtained from 422 trips using logistics robot during single point distribution,and the average time for each trip was 1.75 min.one logistics robot was able to take the workload of 3 to 4 people;logistics robot avoided pedestrians for 420 times;it avoided obstacles for 414 times,it was lost for 13 times,and its speed of operational reaction was normal for 419 times;the comparison results between logistics robot and manual operation showed that T1,P>0.05;T2 and T3,P< 0.01,and the distribution time using robot was less than that of manual operation.Conclusion Using logistics robot for distribution of high value consumables can shorten the time of applying consumables,operation with high distribution accuracy and information precision.

BACKGROUND: Transforming growth factor (TGF)-ß is involved in many physiologic processes, it often promotes metastasis, and its high expression is correlated with poor prognosis. In the present study, we analyzed the correlation between transforming growth factor beta 1 (TGF-ß1) expression and prognosis in intrahepatic cholangiocarcinoma RESULTS: We examined the expression of TGF-ß1 in 78 intrahepatic cholangiocarcinomas by immunohistochemistry and correlated the expression with clinicopathological parameters. TGF-ß1 was expressed in 37 of 78 (47.4 %) intrahepatic cholangiocarcinomas. The expression of TGF-ß1 was significantly correlated with lymph node metastasis, distant metastasis, and tumour recurrence. Patients with TGF-ß1-positive tumours had significantly shorter survival time. In a multivariant analysis, the expression of TGF-ß1 and the tumour stage were independent prognostic factors CONCLUSIONS: Our data suggest that expression of TGF-ß1 is a novel prognostic marker for intrahepatic cholangiocarcinoma.

This research was carried out to examine cytopathological effects of Helicoverpa armigera Cytoplasmic polyhedrosis virus (HaCPV) on infected midgut cotton bollworm (Helicoverpa armigera) using transmission and scanning electron microscope. The symptoms on infected host larvae of the host, compared with healthy ones, were getting swollen with milky-white and fragile Histopathological examinations showed infection with HaCPV small polyhedral inclusion bodies (PIB) after 1 or 2 days which were observed in columnar cells of midgut. Virions were partially or completely occupied in a polyhedral matrix to form polyhedral inclusion bodies (PIB) at periphery of virogenic stroma. PIBs were measured 0.5 to 3.5 mm and virions about 46 nm in diameter. Microvilli of infected columnar cells were affected and degenerated immediately prior to rupture of the cell. Some infected columnar cells ruptured to release PIB into the gut lumen 3 days after infection. In addition,PIB were found in goblet cells, 5 or 6 days after infection. Infected goblet cells degenerate to such an extent that only a few of the original microvillus-like cytoplasmic projections and cell organells were left. These cytopathic effects caused in the midgut by HaCPV on cotton bollworm larvae are essentially similar to those have been reported for lepidoperan and dipteran infection by CPV.

<p><b>OBJECTIVE</b>To investigate the expression of CD95 and special marker for activation of peripheral blood lymphocytes in patients with hand foot and mouth disease (HFMD) and its significance.</p><p><b>METHODS</b>Immunofluorescent two-color flow cytometry was used to study the expression of CD95 and HLA-DR on lymphocytes in 58 patients with HFMD and 34 normal controls.</p><p><b>RESULTS</b>Expression of CD3+ T cells was significantly lower in patients (63.82 +/- 7.74)% than that in controls (P < 0.001), meanwhile the expression of CD4+ T cells was (34.29 +/- 7.33)%, significantly lower than that of the controls (P < 0.005). The percentage of lymphocytes expressing HLA-DR in patients was (23.77 +/- 5.78)%, significantly higher than that of the controls (P < 0.005). Significant difference was observed in the expression of HLA- DR on CD8+ T cells in patients (1.34 +/- 1.12)% as compared with controls (P < 0.005). No significant difference in the expression of CD95 on lymphocytes was observed between patients and the controls (P > 0.05).</p><p><b>CONCLUSION</b>The findings support that cellular immunodeficiency exists in patients and that lymphocytes were abnormally activated in the patients. The activation of peripheral blood T lymphocytes in patients mainly involves CD8 subset and it may play an important role in the immune response to antiviral infection.</p>

In order to develop the heroin abstainers' cue-sensitization questionnaire (HACSQ),and evaluate its reliability and validity,during March and April in 2006,the cross-sectional sample survey and cluster sampling were adopted among 939 volunteer participants who were heroin abstainers from 5 compulsive detoxification centers in China.The data were collected by the self-report ques-tionnaires.The reliability analysis included Cronbach's a coefficient,split reliability and test-retest reliability,while construction validity was analyzed in the light of exploratory and confirmatory fac-tor analysis.Based upon the exploratory factor analysis,4 factors were extracted,which were social emotion cue (SEC),direct implement cue (DIC),personnel cue (PC) and indirect implement cue (IIC),and they together explained 64.52% variance.According to the data,test-retest reliability coefficient and HACSQ.The adjusted goodness of fit index was 0.75.It was suggested that the HACSQ demon-strated good validities and reliabilities,and may be used as a reliable assessment tool for the cue-sensitization among the heroin abstainers.

RESUMO

A L-arginine-producing mutant UN100-12(SG~(r),AE~(r)) was derived from Corynebacterium glutamicum ATCC138761 by combination treatment with ultraviolet(UV) and N-methyl-N-nitro-N-nitrosoguanidine(NTG). It could accumulate 16.6g/L L-arginine in the medium containing glucose as carbon source and ammonium sulfate as nitrogen source, when it was culrured at 30℃ for 4 days. And also this mutant has good stability of descendiblity of L-arginine.
